我的样品有羟基氢和羧基氢等活泼氢,我如果先用氘代甲醇溶解没有发现活泼氢(应该是被交换了),然后蒸干,再用氘代二甲基亚砜溶解。这样能看到活泼氢吗?会不会活泼氢已经以D的形式存在了,而不是H的形式,即使把氘代甲醇蒸干也没有变回H的形式,导致最后用氘代二甲基也无法检测到活泼氢?

yes, vacuumm is crucial for MS coze the gas molecular ion will fly and hit the detector according Newton equation assuming the analyte ion will not change its travel trajectory during its flight. therefore ion shoud be free from collision with other non-analyte ions. the only way to deal with it is to use vacuume to get most of other gas molecules out of chamber of MS. Based on the Bolzemann theory, once the analyte flight distance is shorter than the free-flight-distance, the chance of analyte ion collides with other ions is low.
This is the whole idea why use vaccumm in MS. Man!
