Operation instruction of Agilent1100 HPLC

1.    Open up the computer.
2.    Switch on the power of the detector, the column thermostat, the autosampler, the quaternary pump and the vacuum degasser.
3.    Click “instrument online”, entering the Chemstation.
4.    Purging the pump:
4.1    Manually open the purge valve on the pump,
4.2    In the System Diagram, click on the pump, and select Set up Pump from the menu. Set the Flow to 5 ml/min. set 50% B.(Acetonitrile in channel B)
4.3    Turn on the pump, and leave the pump purging for 10 minutes.
5.    Equilibrating the system: set the Flow to 1 ml/min and 80% B. Watch the online plot; when the baselines have stabilized, leave the system pumping for at least 15 minutes to equilibrate.
6.    Set up or open a method to run the sample.
7.    Before switching the instrument off, flush the system with 100% water for about 20 minutes and then with acetonitrile or methanol for 10 minutes, setting the Flow to 0.4 ml/min.
8.    Exit the Chemstation and shut down the computer.
9.    Switch off the power of the modules.
