Operation Instruction for Waters 2695-2487
1. Switch on the power of the Waters 2695 and the Detector 2487.
2. Perform a wet prime.
2.1 In the Status screen Composition field, enter 100% for the first solvent you want to use for wet priming.
2.2 Press the Direct Functions screen key. Select the Wet Prime option, then press Enter. Enter the flow rate (7.5 ml/min) and the duration time (6 min), then press OK.
2.3 Repeat wet prime steps for each solvent, as appropriate.
3. Purging the Sample Management System.
3.1 In the Status screen, enter the appropriate solvent composition in the Composition fields.
3.2 Press Direct Functions screen key. Select Purge Injector, then press Enter. Enter the number of sample loop volumes (use the default value of 6.0 volumes). Then press Enter. Leave the Compression Check box blank. Press OK.
4. Open up the computer.
5. Click "Empower login", Log in to Empower software with "user name: system; Passwords: manager".
6. Create a new project.
Click "Configure System" in "Empower Pro" window, entering "Configuration Manager" window. Choose "file > new > project" to create a project.
7. Acquire sample data in Run Samples.
7.1 Click "Run Samples" in "Empower Pro" window, choosing the project and entering "Run Samples" window.
7.2 Click "Edit > instrument method" to set up a new instrument method. Click "Edit > new method set", using Winzard to create a new method set. If the method set have already been set up before, then use "open".
7.3 Click "File > new sample set method > using sample set winzard" to create a new sample set method. Or open an existent sample set method.
7.4 Click "monitor" to equilibrate the chromatographic system. Watch the online plot; when the baselines have stabilized, leave the system pumping for at least 15 minutes to equilibrate.
7.5 Click "Injector > run sample set".
8. Process the data and generate the report.
8.1 Click "Browse project" in "Empower Pro" window, choosing the project and entering "Browse project" window.
8.2 In "Channels", choose the sample, double click to review and process the result. Save the process method.
8.3 Come back to "Browse project" window. In "Results", choose the sample, right click to "Preview/publisher". "OK" to create a report.
9. Before switching the instrument off, flush the system with 100% water for about 20 minutes and then with acetonitrile or methanol for 10 minutes, setting the Flow to 0.4 ml/min.
10. Exit the Empower and shut down the computer.
11. Switch off the power of Waters 2695 and Detector 2487.
