High-Performance Liquid Chromatography for the Determination of the Hippuric Acid as Urinary Metabolite of Toluene

Abstract
Objective:To establish a new analytical method for the determination of hippuric acid in human urine by HPLC. Method:Urine samples were collected at the end of the shift and analyzed for hippuric acid by HPLC with a uv/vis detector. A mixed solution of methanol/0.01mol/L phosphoric acid (40/60) as mobile phase was used. The detection was performed at λ=228nm, at 1.0ml/min. Results: The method showed good linearity in a range of 0.01mg/ml～0.5 mg/ml with a correlation coefficient of 0.9999. The average relative standard deviations(RSDs) at three concentration levels of low, medium and high were 2.59%, 0.90%, 0.68% respectively, and the average recovery were 92.25%, 97.27%, 100.15% respectively. The minimum detection concentration was 0.41mg/L. Conclusion:The developed method was simple, reliable, and useful for the biologic monitoring of industrial exposure to toluene with high reproducibility and stability.
