流速:1.000ml/min 柱温:30℃
2.2.2.2 测试方法:
按下表设置梯度洗脱程序,以两通(有阻尼管更好)代替色谱柱连接管路,先用超纯水冲洗系统,待基线平稳后开始执行梯度程序,画出梯度变化曲线。测定三次,取各梯度级高度的平均值,由曲线图测量100%B(C)梯度级平均高度计算各梯级实际体积%,每个梯级的理论体积%和实际体积%之差即为梯度准确度,以最大差值为梯度准确度误差。

2.2.2.3 测试标准:梯度准确度误差≤±1%
网上弄来的,反正意思就是那样,用丙酮的吸收强度来观察梯度变化,丙酮量高时,采集的基线就上升,反之就下降,不一定那设置那么多的梯度。
