我在做免疫分析的时候想通过聚电解质静电吸附方法来固定单克隆抗体,想请教一下小鼠抗人单克隆IgG等电点一般多少?邮箱: lgs005@126.com

普遍地IgG的等电点为8.0,在pH=8的情况下不结合基团。
但是如果考虑到IgG分为4个亚型,每个亚型的等电点又各有不同,分布范围很广,至少从6到9都有
IgG的等电点为pI 8.0,IgM,IgA的等电点为pI 7.0,7.4,在pH 8.0时IgG带正电荷 
I found the heavy chain sequence of RAT IgG2 and the light chain sequence of MOUSE kappa chain and the predicted PI is 7.2. This is how you calculate the PI of a protein. If I put in MOUSE lambda light chain sequence, the predicted PI is 8.8.

I did not dig deeply but I did not find the light chain sequence of rat light chain. Mouse and rat light chain should have very similar sequence.

Try to see if you can find the correct sequences.

好像多少的都有。
单抗应该好办,因为是单一蛋白了。
预先测定一下,就OK了。
